Determination of Dissociation Constant and Binding Constant of Famciclovir

Abstract: Both the dissociation constants of famciclovir obtained were 3.8 using ultraviolet spectrophotometry with the wavelengths at 225 nm and 305 nm. Bovine serum albumin was added to a Na2HPO4-KH2PO4 buffer solution (pH 7.0) with famciclovir. The binding constant was measurd by the double-reciprocal analysis. The results showed that the bovine serum albumin and famciclovir formed a 1:4 complex, and the binding constant obtained was 4.51×106L/mol. This method is simple and reliable.
