Abstract: The glycosylation catalyzed by β-glucosidase for synthesis of 4-hydroxyl phenylethyl β-D-glucoside (salidroside) in mixed medium of ionic liquids/organic solvent/buffer solution was studied. The effects of some important factors on the glycosylation such as the constitution of the medium, and the reaction conditions were investigated. The results showed that 1-butyl-3-methylimidazolium hexafluorophosphate (C4MIm·PF6) /1,4-dioxane/citric acid-phosphate buffer was the proper mixed medium for biosynthesis of salidroside. When the volume proportions of these components were 17%, 68% and 15% respectively, the product concentration would reach 18.9 mg/mL under these conditions: D-(+)-glucose 100 mg/mL, 4-hydroxyphenethyl alcohol (tyrosol) 220 mg/mL, β-glucosidase 12 U/mL, pH 6.0, 50 ℃ and 275 rpm for 120 h.

Key words: β-Glucosidase; salidroside; ionic liquid; glycosylation
